Automated solution-phase multiplicative synthesis of complex glycans up to a 1,080-mer

Carbohydrates play essential roles in nature, such as cell–cell communication, cell growth and immunoresponse. However, the synthesis of structurally well-defined carbohydrates, especially large-sized glycans, is a challenging task. Here we report an automated solution-phase multiplicative complex glycans enabled by preactivation-based, multicomponent, one-pot glycosylation continuous multiplying amplification. This was achieved making dual-mode glycan synthesizer. Using this synthesizer, library oligosaccharides covering various glycoforms glycosidic linkages assembled rapidly, either general promoter-activation mode or light-induced-activation mode. The fully protected fondaparinux pentasaccharide (an anticoagulant) realized on gram scale. Furthermore, ten-component tandem reactions were performed, allowing assembly arabinans up to 1,080-mer using strategy. complexity carbohydrate structures makes their challenging. Now, synthesizer reported which capable preparing bioactive oligosaccharides, including pentasaccharide. can rapidly assemble size, starting from monosaccharide building blocks.
